The present study characterized the homologous and heterologous immune response in type-I porcine reproductive and respiratory syndrome virus (PRRSV) infection. Two experiments were conducted: in experiment 1, eight pigs were inoculated with PRRSV strain 3262 and 84 days post-inoculation (dpi) they were challenged with either strain 3262 or strain 3267 and followed for the next 14 days (98 dpi). In experiment 2, eight pigs were inoculated with strain 3267 and challenged at 84 dpi as above. Clinical course, viremia, humoral response (neutralizing and non-neutralizing antibodies, NA) and virus-specific IFN-γ responses (ELISPOT) were evaluated all throughout the study. Serum levels of IL-1, IL-6, IL-8, TNF-α and TGF-β were determined (ELISA) after the second challenge. In experiment 1 primo-inoculation with strain 3262 induced viremia of ≤ 28 days, low titres of homologous NA but strong IFN-γ responses. In contrast, strain 3267 induced longer viremias (up to 56 days), higher NA titres (≤ 6 log2) and lower IFN-γ responses. Inoculation with 3267 produced higher serum IL-8 levels. After the re-challenge at 84 dpi, pigs in experiment 1 developed mostly a one week viremia regardless of the strain used. In experiment 2, neither the homologous nor the heterologous challenge resulted in detectable viremia although PRRSV was present in tonsils of some animals. Homologous re-inoculation with 3267 produced elevated TGF-β levels in serum for 7–14 days but this did not occur with the heterologous re-inoculation. In conclusion, inoculation with different PRRSV strains result in different virological and immunological outcomes and in different degrees of homologous and heterologous protection.  相似文献

The necessity, and in particular the timing, of gonadectomy in dogs and cats is still controversial. This is mainly because gonadectomy confers a mixture of benefits and adverse effects that depend upon the age at neutering, sex, species and breed. In this paper, the long-term risks and benefits of gonadectomy, at various ages, in dogs and cats are reviewed using data describing the effects of desexing on the urogenital tract and on other medical conditions, such as orthopaedic diseases, immune-mediated diseases, tumour development and behaviour.  相似文献

Nitrogen emissions have grown in Spain during the last 15 years. As precipitation scavenges gases and aerosols from the atmosphere, an effect on rainwater concentrations can be expected. However, time-series studies on wet N concentrations in the Iberian Peninsula are very scarce. This paper aims to fill this gap by analysing weekly rainfall N concentrations at a set of rural sites in Catalonia (NE Spain) from 1995/1996 to 2007 and a forest site monitored from 1983 to 2007. The sites encompass a range of rural environments and climate conditions, from the inland pre-Pyrenees (Sort) to the Mediterranean coast (Begur) and from north (Sort and Begur) to central (Palautordera and La Castanya) and south Catalonia (La Senia). We found a 1-year cycle for concentrations of NH 4 + and NO 3 ? whereby higher values were reached at the end of spring–early summer, except at the easternmost coastal site of Begur. Weekly NH 4 + concentrations decreased with time at all sites (except at La Senia) whilst NO 3 ? concentrations increased at all sites during the same period. Rainfall SO 4 2? concentrations decreased with time at all sites. The opposite trends in NO 3 ? and SO 4 2? concentrations determined a shift in the relative acid contribution of those anions during the 12–13-year period. To interpret the increasing trend, mean annual NO 3 ? concentrations were regressed against NO2 Spanish emissions and to some indicators of local anthropogenic activity. The increase at Sort and Palautordera showed good correlation with local anthropogenic indicators. Wet inorganic N deposition ranged between 4.2 and 6.7 kg ha?1 year?1. When including estimates of dry deposition, total annual deposition rose up to 10–20 kg ha?1 year?1, values that have been found to initiate adverse effects on Mediterranean-type forest ecosystems.  相似文献

Irrigation with saline water is one of the major problems in citrus crop in arid and semi-arid regions. Because rootstock and fertilization play an important role in citrus salt tolerance, we investigated the influence of the nitrogen fertilization and rootstock on salt tolerance of 2-year-old potted Fino 49 lemon trees. For that, trees grafted on Citrus macrophylla (M) or Sour orange (SO) rootstocks were watered for 12 weeks with complete nutrient solution containing either 0 mM NaCl (control, C), 50 mM NaCl (S), 50 mM NaCl with an additional 10 mM potassium nitrate (S + N), or 50 mM NaCl with a 1% KNO3 (S + Nf) foliar spray application. Trees on M were more vigorous than trees on SO and saline treatments reduced leaf growth similarly in trees on both rootstocks. Trees on SO had a lower leaf Cl and Na+ concentration than those on M. Additional soil nitrogen (S + N) decreased leaf Cl concentration and increased leaf K+ concentration in salinized trees on both rootstocks. However, the salinity-induced reduction leaf growth was similar in S + N and S trees. This was due to osmotic effect, beside leaf Cl and Na+ toxicity, played an important role in the growth response of Fino 49 lemon to the salt stress. Additional foliar nitrogen in the S + Nf treatment also reduced leaf Cl concentration relative to the S treatment but trees from S + Nf treatment had the lowest leaf growth. Net assimilation of CO2 (ACO2ACO2), stomatal conductance (gs) and plant transpiration were reduced similarly in all three salt treatments, regardless rootstock. Salinity reduced leaf water and osmotic potential such that leaf turgor was increased. Thus, the salinity-induced ACO2ACO2 reductions were not due to loss of turgor but rather due to high salt ion accumulation in leaves.  相似文献

A chronological study of central nervous system disorders induced by Marek's disease virus (MDV) has been conducted. Neurologic clinical signs were recorded daily for individual chickens of two genetic lines after inoculation of 13 serotype 1 MDV strains representing all three pathotypes. In addition to classical transient paralysis (TP) previously described by many workers, and acute TP, described in the companion paper, we have identified for the first time two other neurologic syndromes, persistent neurologic disease (PND) and late paralysis (LP). PND designates birds that showed a variety of neurologic signs (ataxia, torticollis, and nervous tics) after recovery from paralysis (12-15 days postin-oculation [DPI]) that either persisted through the observation period or presented a cyclic pattern. LP was a rare syndrome characterized by the late onset of the paralytic stage (about 20 DPI), perhaps indicating occasional failure of the initial intraabdominal inoculation to induce infection. Clinical signs and histopathologic alterations of the brain were also evaluated sequentially in chickens of two genetic lines after inoculation with two MDV strains (virulent MDV and very virulent plus MDV). Although clinical response differed greatly among treatment groups, types of lesions (endotheliosis, mononuclear perivascular cuffing, vasculitis, vacuolization, and increase in cellularity of the neuropil) were similar. However, early onset of lesions (by 6 days) appeared to be associated with a greater severity of clinical signs. We also found that neurologic response was greatly influenced by viral pathotype (virulence). This study thus confirms that the central nervous system is an important target organ for MDV resulting in several distinct clinical manifestations and suggests that neurologic responses in antibody-free chickens might be a useful criterion for virus pathotyping.  相似文献

REASONS FOR PERFORMING STUDY: This study reports a neurological disease unrecognised until now in ponies in southern Brazil. HYPOTHESIS: Epidemiological data strongly suggests that the ingestion of Sida carpinifolia is involved in the aetiology. We tested the hypothesis that it is an acquired lyosomal storage disease. METHODS: Following the death of 3 ponies, all ponies from the premises were closely monitored; epidemiological data and clinical findings carefully recorded. Fragments of several organs, including CNS, were fixed in neutral formalin and embedded in paraffin-wax. Sections were stained with haematoxylin and eosin. Representative sections of the cerebellum and trigeminal ganglia were submitted to lectin histochemical procedures. RESULTS: The neurological disorder, characterised by stiff gait, muscle tremors, abdominal pain and death, was observed on a farm with 3 hectares of pasture. Three of 11 ponies died 15-20 days after they had been introduced into a new paddock heavily infested by the plant Sida carpinifolia. No significant gross lesions were observed. The main histological findings included multiple cytoplasmatic vacuoles in swollen neurones in the brain, cerebellum, spinal cord, autonomic ganglia (trigeminal and celiac ganglia), and submucosal and myenteric plexus of the intestines. In the kidneys, there was marked vacuolation of the proximal convoluted tubular cells. Sections of cerebellum and trigeminal ganglion were submitted to lectin histochemistry. The vacuoles in different cerebellar and ganglion cells reacted strongly to the following lectins: Concanavalia ensiformis, Triticum vulgaris and succinylated-Triticum vulgaris. CONCLUSIONS: The pattern of staining coincides with that of both swainsonine toxicosis and inherited mannosidosis reports. The histopathological changes were similar to those described in S. carpinifolia spontaneous and experimental poisoning in goats. This disease seems to be similar to Swainsona, Oxytropis and Astragalus toxicosis. POTENTIAL RELEVANCE: S. carpinifolia should be evaluated as a possible cause in the diagnosis of equine neuropathies.  相似文献

The proportions of fibres of the collagenous and elastic systems were measured in the aortas of three normal heifers and in nine heifers given the calcinogenic plant Solanum glaucophyllum for 15, 30 or 60 days. There were decreases in the amount of collagen relative to reticulin, and in the proportion of elastic fibres which were related to the period of dosing. These changes may have an adverse influence on the animals' cardiovascular function.  相似文献

The presence of annulate lamellae in the germ cells of the chick embryo, in stage 9 of H amburger /H amilton (1951), as well as in gonadal germ cells of male chick embryos which had been incubated for 81/2 days, both in normal and experimental conditions, suggests that these formations may be associated with some specific function in the development of this cell line.  相似文献

Fire affects large parts of the dry Mediterranean shrubland, resulting in erosion and losses of plant nutrients. We have attempted to measure these effects experimentally on a calcareous hillside representative of such shrubland. Experimental fires were made on plots (4 m × 20 m) in which the fuel was controlled to obtain two different fire intensities giving means of soil surface temperature of 439°C and 232°C with temperatures exceeding 100°C lasting for 36 min and 17 min. The immediate and subsequent changes induced by fire on the soil's organic matter content and other soil chemical properties were evaluated, together with the impact of water erosion. Seven erosive rain events, which occurred after the experimental fires (from August 1995 to December 1996), were selected, and on them runoff and sediment produced from each plot were measured. The sediments collected were weighed and analysed. Taking into account the variations induced by fire on the soil properties and their losses by water erosion, estimates of the net inputs and outputs of the soil system were made. Results show that the greatest losses of both soil and nutrients took place in the 4 months immediately after the fire. Plots affected by the most intense fire showed greater losses of soil (4077 kg ha?1) than those with moderate fire intensity (3280 kg ha?1). The unburned plots produced the least sediment (72.8 kg ha?1). Organic matter and nutrient losses by water erosion were related to the degree of fire intensity. However, the largest losses of N‐NH4+ and N‐NO3 by water erosion corresponded to the moderate fire (8.1 and 7.5 mg N m?2, respectively).  相似文献

Apoptosis in the testis is required to ensure an efficient spermatogenesis. However, sometimes, defective germ cells that are marked for elimination during this process escape elimination in the testes, giving rise to ejaculates with increased percentages of abnormal and apoptotic spermatozoa and a high percentage of apoptotic bodies. Apoptosis markers in the ejaculate have been associated with low fertility, either in animals or humans. Therefore, the goal of this study was to investigate whether fresh equine semen contains apoptotic bodies [initially named Merocyanine 540 (M540) bodies] and to study the relationship between the quantity of these bodies and cell concentration, the volume of ejaculate, viability and motility. Moreover, we also studied whether the presence apoptotic bodies in fresh semen was related to the resistance of the stallion spermatozoa to being incubated at 37°C or being frozen and thawed. Fresh equine semen was stained with fluorescent dyes such as M540 and Annexin‐V. Active Caspase 3 was studied in fresh semen through Western blotting and immunofluorescence with a specific antibody. Sperm kinematics was assessed in fresh, incubated and thawed samples using computer‐assisted semen analysis, and viability was evaluated with the LIVE/DEAD Sperm Viability Kit. Overall, our results demonstrate for the first time the presence of apoptotic bodies in equine semen. The quantity of apoptotic bodies was highly variable among stallions and was positively correlated with Caspase 3 activity in fresh samples and negatively correlated with the viability and motility of stallion spermatozoa after the cryopreservation process.  相似文献
